1.科技領域
自身免疫甲狀腺疾病(Autoimmune thyroid diseases,AITDs)包括Graves病(Graves’ disease👨🏽💼, GD)和橋本甲狀腺炎(Hashimoto’s thyroiditis,HT)🕯,其確切發病機製尚不明確🖋,本項目依托熊猫体育官网,結合基礎、臨床研究,歷經12年從AITDSs的遺傳學和表觀遺傳學發病機製展開研究,主要從基因多態性(Single nucleotide polymorphism, SNP)、微小RNA(microRNA)、DNA基因甲基化、組蛋白乙酰化等方面著手,多方面🐤、多角度地對AITDSs的遺傳學和表觀遺傳學發病機製進行了深入的探討和研究☂️。
2.主要技術創新內容
1)檢測了兩個甲狀腺自身抗原基因和多個免疫相關基因的單核苷酸多態性與AITDs發病之間的關系🧚♂️,並發現甲狀腺球蛋白(Thyroglobulin, Tg)基因與TSH受體(TSHR)基因是中國漢族人群AITDs發病的易感基因;發現以下24個免疫相關基因CD40、CTLA-4、IL-17👈🛎、IL-21、IL-21R、ST2、IL18R1、IL18RAP、CLEC16A🕵🏻、TNFAIP3、STAT4、 IRAK1🐈🛁、MECP2🧏🏽🏄🏽♀️、TRAF1🫰🏽、RNASET2🏂🫃🏻、UBE2L3🧛🏿、VDR、TNFRSF1A⛷🧑🦼、IKZF3、IRGM、BANK1🕺🏽、TNFSF4👈🏽、STAT3和IL-22可以增加AITDs的發病風險🤛🏻。本項目研究的中國漢族人群多個免疫相關基因的多態性與AITDs的關系🧝🏽♀️,在國內外屬首次報道,故本系列研究屬原創性研究🚴🏼♀️,具有較好的創新性和先進性。
2)初步建立了系統的、特異的GD疾病差異拷貝數變異(Copy number variation, CNV)表達譜🫲🏻🤽🏼。後續對芯片篩選出的部分差異表達CNV基因進行驗證⛽️,同時對CNV差異表達基因TRY6相關的兩個SNP位點進行檢測👐🏽✯,結果發現CFH🍴、CFHR1🛌🏻、KIAA0125、UGT2B15、UGT2B17、TRY6、CCL3L1拷貝數與GD發病風險無關。
3)運用表達人TSH受體(TSHR)A亞單位的腺病毒載體免疫近交系BALB/c小鼠🧗🏻♀️,成功製備了具有人類GD主要特征的動物模型🧚🏿♂️。
4)首次發現甲狀腺局部的CTLA4Ig基因的註射可通過誘導Thl優勢向Th2偏移而對自身免疫性甲狀腺炎起治療作用。
5)首次發現AITDs的甲狀腺組織中存在異常表達的microRNAs;外周血單個核細胞的DNA甲基化、組蛋白乙酰化表達異常與GD發病存在相關性。
3、知識產權情況
本項目受三項國家自然科學基金(30871185🌸🫔、81070627、81270871)資助。本研究形成主要論文20篇,其中SCI收錄19篇🎁,總被引用次數達284次🧑🏿🎄。同時還參加了多次熊猫体育交流包括在全國和上海的內分泌年會上作口頭或書面交流並獲獎🧖🏿♀️;共培養碩士🫶🏻、博士研究生40余名。
該研究成果主要在甲狀腺疾病的研究和臨床領域內推廣和應用,有助於加深內分泌醫生對AITDs發生機理的理解和認識⏭,為AITDs的精準治療、特異性免疫治療提供依據和思路👩🏿🔧,具有較好的臨床應用前景。
